Oil bodies were purified from 4-d-germinated cotyledons by discontinuous Suc gradient centrifugation. After centrifugation, samples were taken from the bottom, middle, and top layers and served as 20%, 15%, and 8% fraction protein and enzyme sources, respectively. The white pad on top of the gradient was collected and used as unwashed oil bodies. Unwashed oil bodies were gently resuspended in 15% (w/v) Suc in 20 mm Tricine buffer at pH 7.5, subjected to one additional centrifugation, and used as washed oil bodies. LOX activity was assayed by measuring O2consumption. The activities of the crude samples were taken as the total activity. Relative LOX activity is equal to the activity in the fraction divided by the activity of the crude samples. Values are means ± se of three independent experiments.
